No matter how you measure it what you want to achieve is that through the suspension travel the chain does not get tight. The most common cause for chair wear is a tight chain.

So the way I do this is to sit on the bike and feel how much slack I have. So long as I can feel enough movement to just not touch the bottom of the swingarm then that's what I want.

A slightly loose chain is better that a chain to tight.
